| ¹ß¾Æ½ÃŲ ¹°¿À¸®³ª¹«(Alnus hirsuta)ÀÇ ÇϹèÃà ºÎÀ§¸¦ Àý´ÜÇÏ¿© ¾òÀº ¶±ÀÙÀÌ Æ÷ÇÔµÈ »óºÎ¸¦ 6% glucose, MS(Murashige and Skoog, 1962) ºñŸ¹Î È¥ÇÕü, PVP(polyvinylpyrrolidone) ¹× $0-50;mu extrm{M}$ÀÌ BAP(6-benzylaminopurine)¸¦ ÷°¡ÇÑ NT(Nagata and Takebe, 1971) ¹«±â¿°¹èÁö¿¡¼ ¹è¾çÇÑ °á°ú, $5;mu extrm{M}$ÀÌ BAP°¡ shoot Áõ½Ä¿¡ °¡Àå ÀûÇÕÇÑ °ÍÀ¸·Î ³ªÅ¸³µ´Ù. µû¶ó¼ ÀÌ ¹èÁö¿¡¼ shoot Áõ½ÄÀ» °è¼ÓÇÏ¿© ½ÇÇè¿¡ »ç¿ëÇÒ 200¿©°³ÀÇ shoot¸¦ È®¸³ÇÏ¿´´Ù. Shoot Áõ½Ä¿¡ ¹ÌÄ¡´Â ¹«±â¿°·ù¿Í ź¼Ò¿øÀÇ ¿µÇâÀ» Á¶»çÇÑ °á°ú ź¼Ò¿ø¿¡ ¹«°üÇÏ°Ô NT ¹«±â¿°ÀÌ MS ¹«±â¿°¿¡ ºñÇØ 3-12¹èÀÇ Áõ½ÄÀ» ³ªÅ¸³ÂÀ¸³ª NT ¹«±â¿°¹èÁö¿¡¼ ź¼Ò¿øÀº Ưº°ÇÑ Â÷À̸¦ º¸¿©ÁÖÁö ¾Ê¾Ò´Ù. ¹ß±Ù¿¡ ¹ÌÄ¡´Â NT ¹«±â¿°, sucrose, IBAÀÇ ³óµµ¿¡ µû¸¥ ¿µÇâÀ» Á¶»çÇÑ °á°ú 1/4-1/2 ³óµµÀÇ NT ¹«±â¿°, 3%ÀÇ sucrose, $1-8;mu extrm{M}$ÀÇ IBA¿¡¼ 100% °¡±î¿î ¹ß±ÙÀ²À» º¸¿´À¸¸ç »Ñ¸®¹ß´Þµµ ¾çÈ£ÇÏ¿´´Ù. ¹ß±ÙµÈ °³Ã¼¸¦ Åä¾ç¿¡ À̽ÄÇÏ°í ½Àµµ¸¦ Á¡ÁøÀûÀ¸·Î ³·Ãß¸é¼ ÀûÀÀ½ÃŰ¸é °ÅÀÇ 100%ÀÇ »ýÁ¸À²À» ³ªÅ¸³»¾ú´Ù. |

| Shoot tip explants from germinated seeds of Alnus hirsuta were cultured on NT (Nagata and Takebe, 1971) mineral salts medium supplemented with 6% glucose, MS (Murashige and Skoog, 1962) vitamin mixture, polyvinylpyrrolidone (PVP) and $0-50;mu extrm{M}$ 6-benzylaminopurine (BAP). Five $mu extrm{M}$ BAP was found to give the highest shoot multiplication rate. Accordingly about 200 shoots were obtained for further experiments by multiplying shoots on this medium for 4-5 months. Regardless of carbon sources, NT mineral medium produced 3-12 times of shoots than MS mineral medium did. On NT mineral medium, 3% sucrose, 3% glucose and 6% glucose yielded no significant differences. It was observed that media consisting of 1/4-1/2 strength NT mineral salts, 3% sucrose and $1-8;mu extrm{M}$ IBA produced about 100% rooting rate. Almost 100% of the resulting plantlets survived after transfer to the soil by decreasing humidity stepwise.epwise. |
