| º» ¿¬±¸ÀÇ ¸ñÀûÀº ¾çÆÄÃßÃâ¹°ÀÇ °£º¸È£ ¹× Ç×»êÈ È¿°ú¸¦ Á¶»çÇϱâ À§ÇÔÀÌ´Ù. °£ ¼Õ»óÀ» À¯¹ß½ÃŰ´Â t-BHP (1.5 mM) Á¸ÀçÇÏ¿¡ °£¼¼Æ÷¸¦ 0, 0.01, 0.05, 0.1 ¹× 0.3 mg/mlÀÇ ´Ù¾çÇÑ ³óµµÀÇ ¾çÆÄÃßÃâ¹°·Î 1½Ã°£ µ¿¾È ÀÏÂ÷¹è¾çÇÏ¿´´Ù. t-BHP´Â GOT¿Í LDH Ȱ¼º ¹× TBARS ³óµµ¸¦ Áõ°¡½ÃÄ×À¸¸ç MTT°ªÀº °¨¼Ò½ÃÄ×´Ù. 0.05 mg/ml ³óµµÀÇ ¾çÆÄÃßÃâ¹° ÷°¡´Â t-BHP¿¡ ÀÇÇØ Áõ°¡µÈ GOT ¹× LDH Ȱ¼ºÀ» °¨¼Ò½ÃÄ×À¸¸ç 0.1 mg/ml ³óµµÀÇ ¾çÆÄÃßÃâ¹°Àº t-BHP¿¡ ÀÇÇØ °¨¼ÒµÈ MTT °ªÀ» Áõ°¡½ÃÄ×´Ù. ¶ÇÇÑ 0.01 mg/ml ³óµµÀÇ ¾çÆÄÃßÃâ¹° ÷°¡´Â t-BHP¿¡ ÀÇÇØ Áõ°¡µÈ TBARS ³óµµ¸¦ °¨¼Ò½ÃÄÑ ¾çÆÄÃßÃâ¹°ÀÌ t-BHP¿¡ ÀÇÇØ À¯¹ßµÈ °£¼Õ»ó°ú ÁöÁú°ú»êȸ¦ ¾ïÁ¦½ÃÄ×´Ù. t-BHP 󸮴 °£¼¼Æ÷ÀÇ catalase, GSH-Px ¹× GSH-Rd Ȱ¼ºÀ» ÇöÀúÈ÷ °¨¼Ò½ÃÄ×´Ù. ±×·¯³ª 0.1 mg/ml ³óµµÀÇ ¾çÆÄÃßÃâ¹° ÷°¡´Â t-BHP¿¡ ÀÇÇØ °¨¼ÒµÈ catalase GSH-Px ¹× GSH-Rd Ȱ¼ºÀ» Áõ°¡½ÃÄ×À¸¸ç ƯÈ÷ catalase Ȱ¼ºÀº t-BHP ¹«Ã·°¡±º ¼öÁرîÁö Áõ°¡½ÃÄ×´Ù. ¶ÇÇÑ hydroxyl radicalÀ» »ý¼ºÇÏ´Â Fenton ½Ã¾àÀÇ Á¸ÀçÇÏ¿¡ plasmid DNA¸¦ ¾çÆÄÃßÃâ¹°°ú ÇÔ²² ¹è¾çÇÑ °á°ú ¾çÆÄÃßÃâ¹°Àº ³óµµ ÀÇÁ¸ÀûÀ¸·Î hydroxyl radical¿¡ ÀÇÇØ À¯µµµÈ single-strand Àý´ÜÀ» ¾ïÁ¦ÇÏ¿´´Ù. ÀÌ»ó°ú °°ÀÌ °£¼¼Æ÷ ÀÏÂ÷¹è¾ç¿¡¼ ¾çÆÄÃßÃâ¹°Àº t-BHP¿¡ ÀÇÇØ À¯¹ßµÈ °£µ¶¼º, °£¼¼Æ÷ »ýÁ¸À² °¨¼Ò, ÁöÁú°ú»êȸ¦ ³óµµ ÀÇÁ¸ÀûÀ¸·Î ¾ïÁ¦½ÃÄ×°í ¶ÇÇÑ t-BHP¿¡ ÀÇÇØ ¾ïÁ¦µÈ GSH-Px, GSH-Rd ¹× catalaseÀÇ È°¼ºÀ» Áõ°¡½ÃÄ×´Ù. | The objective of present study was to investigate the hepatoprotective and antioxidative effects of onion extracts. Primary cultures of rat hepatocytes were incubated with 1.5 mM tort-butyl hydroperoxide(t-BHP), potent oxidizing agent to liver, for 1 hr in the presence or absence of various concentrations (0, 0.01, 0.05, 0.1 or 0.3 mg/ml) of onion extract. Incubation with t-BHP increased glutamic oxaloacetic transaminase(GOT) and lactate dehydrogenase(LDH) acitivities and thiobarbituric acid reactive substances(TBARS) concentration but decreased 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) reduction. Onion extracts at the concentration of 0.05 mg/ml decreased t-BHP-induced GOT and LDH activities. Onion extract at the concentration of 0.1 mg/ml increased t-BHP-induced MTT reduction. Onion extract at the concentration of 0.01 mg/ml decreased t-BHP-induced TBARS concentration. Taken together, onion extracts prevented t-BHP-induced hepatocyte injury and lipid peroxidation. Catalase, glutathione peroxidase(GSH-Px) and glutathione reductase(GSH-Rd) activities of hepatocytes were significantly decreased by t-BHP. Onion extracts at the concentration of 0.1 mg/ml prevented t-BHP-induced decrease in catalase, GSH-Px and GSH-Rd activities. Onion extracts prevented hydroxyl radical-induced single-strand breakage in dose-dependent manner when plasmid DNA was incubated with various concentrations of onion extracts in the presence of Fenton reagents producing hydroxyl radical. These results demonstrate that onion extracts suppressed t-BHP-induced cytoctoxicity, decreased viability and lipid peroxidation and increased GSH-Px, GSH-Rd and catalase activities. Thus hepatoprotective and antioxidant effects of onion extract seem to be due to, at least in part, the increase in antioxidant enzyme activities as well as prevention from hydroxyl radical-induced oxidation, followed by inhibition of lipid peroxidation. |
