| Â÷°¡¹ö¼¸ ÀΰøÀç¹è¸¦ À§ÇÑ ±Õ»ç¹è¾çÀû Ư¼ºÀ» Á¶»çÇÑ °á°ú´Â ´ÙÀ½°ú °°´Ù. 1) Â÷°¡¹ö¼¸±ÕÀÇ ÀûÁ¤¹èÁö´Â BDA(Birch Dextrose Agar)¿¡¼ 66.3mm/10ÀÏ·Î ±Õ»ç»ýÀå°ú ±Õ»ç¹Ðµµ°¡ °¡Àå ÁÁ¾ÒÀ¸¸ç, ±× ´ÙÀ½Àº GDA, PDA, CDA, PODA, ODA, YM, MCM, MEA(pH7.2), CHA, MEA(pH4.7) ¼øÀ̾ú´Ù. 2) Â÷°¡¹ö¼¸ÀÇ ±Õ»ç»ýÀå°ú ¹Ðµµ¿¡ °¡Àå ÀûÁ¤ÇÑ ¿Âµµ´Â 3$0^{circ}C$ À̾úÀ¸¸ç 4$0^{circ}C$¿¡¼´Â ±Õ»ç°¡ »ç¸êÇÏ¿´´Ù. KNAC3005 ±ÕÁÖ´Â 3$0^{circ}C$¿¡¼ 66.3mm/10ÀÏ·Î ±Õ»ç»ýÀå°ú ¹Ðµµ°¡ °¡Àå ¾çÈ£ÇÏ¿´À¸¸ç, ¿Âµµº°·Î´Â 25, 20, 15, 35, 10, 5$^{circ}C$¼øÀ¸·Î ±Õ»ç»ýÀå ¼Óµµ¿Í ¹Ðµµ°¡ ÁÁ¾Ò´Ù. 3) Â÷°¡¹ö¼¸ÀÇ ±Õ»ç»ýÀå°ú ¹Ðµµ¿¡ °¡Àå ÀûÁ¤ÇÑ »êµµ(pH)´Â 6.0¿¡¼ 88.4mm/10ÀÏÀÌ¸ç ±×º¸´Ù ³ô°Å³ª ³·À¸¸é ±Õ»ç»ýÀå°ú ¹Ðµµ¿¡ ÀúÇØ¸¦ ¹Þ´Â´Ù. 4) Â÷°¡¹ö¼¸ÀÇ ±Õ»ç»ýÀå¿¡ ÀûÇÕÇÑ Åº¼Ò¿øÀº maltose·Î 331mg/25$mell$/15ÀÏÀ̸ç, ÃÖÀû Áú¼Ò¿øÀº peptoneÀ¸·Î 347mgÀÌ´Ù. ÃÖÀû À¯±â»êÀº glutamic acid·Î 357mgÀ̾ú´Ù ÃÖÀû ºñŸ¹ÎÀº biotinÀ¸·Î 370mg/15daysÀ̸ç C/NÀ²Àº 40ÀÌ ÀûÁ¤ÇÏ¿´´Ù. |

| The results of examining characteristics of mycelial growth and culture condition for determining the condition of artificially culturing Inonotus obliqua (chaga) are as follows. 1) Mycelial growth and density oft, oblique were the highest in the medium of BDA (Birch Dextrose Agar; 66.3mm/10d) followed by the order of GDA, PDA, CDA, PODA, ODA, YM, MCM, MEA (pH 7.0), CHA, and MEA (pH 4.7). 2) Optimal temperature for the mycelial growth and density of 1. obliqua were shown to be 30$^{circ}C$, but the mycelia were dead at 40$^{circ}C$. the mycelial growth and density of KNAC3005 strains was the highest at 30$^{circ}C$(66.3mm/10d) followed by the order of 25, 20, 15, 35, 10, and 5$^{circ}C$. 3) Optimal pH for the mycelial growth and density were revealed to be 6.0 (88.4mm/10d). Above or below pH 6.0, the mycelial growth and density were shown to be retarded. 4) Optimal carbon, nitrogen and organic acid sources for the spawn growth of 40 were maltose (331mg/25$mell$/15d), peptone (347mg/25$mell$/15d), and glutamic acid (357mg/25$mell$/15d), respectively. Optimal level of biotin was 370mg/15d and optimal C/N ratio was 40. |
