AK-07ÀÇ Æ¯¼º°ú 16S rDNAÀÇ ¿°±â ¼¿ ºÐ¼®À» ±âÃÊ·Î ÇÏ¿© À¯¿¬°ü°è¸¦ Á¶»çÇÑ °á°ú, ÇüÅÂÀû, »ý¸®Àû »ýÈÇÐÀû Ư¡µéÀº Acinetobacter¼ÓÀÇ Æ¯¼ºµé°ú À¯»çÇÏ¿´À¸¸ç, 16S rDNAÀÇ ¿°±â ¼¿ ºÐ¼®ÇÑ °á°ú´Â Acinetobacter johnsonii¿Í 99.5%ÀÇ À¯»ç¼ºÀ» ³ªÅ¸³»¾î, Acinetobacter johnsonii AK-07·Î ¸í¸íÇÏ¿´´Ù. ³²Á¶·ù ºÐÇØ Ư¼ºÀ» Á¶»çÇϱâ À§Çؼ, AK-07¸¦ A. cylindrica¿Í È¥ÇÕ ¹è¾ç½Ã Á¢Á¾ 2ÀÏ ÈÄ¿¡ ³²Á¶·ùÀÇ ºÐÇØ°¡ °üÂûµÇ¾ú°í, Á¢Á¾ 10ÀÏ ÈÄ¿¡´Â ³²Á¶·ù°¡ ¿ÏÀüÈ÷ »ç¸êÇÏ¿´À¸¸ç, AK-07ÀÇ ¼¼Æ÷ ¼ö´Â $8{ imes}10^8cfu ml^{-1}$±îÁö Áõ°¡ÇÏ¿´´Ù. ±×·¯³ª ¹è¾ç »óµî¾×À» A. cylindrica¿Í È¥ÇÕ ¹è¾ç ÇÏ¿´À» ¶§¿¡´Â ³²Á¶·ùÀÇ ºÐÇØ´Â °üÂû µÇÁö ¾Ê¾Ò´Ù. µû¶ó¼ AK-07´Â ³²Á¶·ù¸¦ Á÷Á¢ Á¢ÃËÇÏ¿© ºÐÇØÇÏ´Â °ÍÀ¸·Î »ç·áµÇ¾î, AK-07¿¡ ¼¼Æ÷¿¡ Á¸ÀçÇÏ´Â È¿¼ÒÀÇ È°¼ºÀ» Á¶»çÇÑ °á°ú Protease¿Í glycanases Áß¿¡¼ ¥â-XylosidaseÀÇ È°¼ºÀÌ °¡Àå ³ô¾ÒÀ¸¸ç, Alginase, Laminarinase, Lipase, ¥â-Galactosidase ¹× ¥â-GlucosidaseÀÇ È°¼ºµµ ³ôÀº ¼öÁØÀ¸·Î °üÂûµÇ¾ú´Ù. A. johnsonii AK-07Àº A. cylindricaÀÇ polysaccharides³ª peptidoglycans¸¦ monosaccharidesÀ̳ª ÀúºÐÀÚ À¯±â¹°·Î ºÐÇØÇÏ´Â °ÍÀ¸·Î ¿©°ÜÁø´Ù. |

| To investigate bacteria with algal lytic activities against Anabaena cylindrica when water blooming occurs and to study enzyme profiles of alga-lytic bacteria, various bacterial strains were isolated from surface waters and sediments in eutrophic lakes or reservoirs in Korea. A bacterial strain AK-07 was characterized and identified as Acinetobacter johnsonii based on its 16S rDNA base sequence. When AK-07 was cocultivated with A. cylindrica, bacterial cells propagated to $8{ imes}10^8cfu ml^{-1}$ and lyses algal cells. However, culture filtrates of AK-07 did not exhibit algal lytic activities. That suggesting the enzymes on the surfaces of the bacterium might be effective algal lytic agents to cause lyses of cells. Acinetobacter johnsonii AK-07 exhibited high degradation activities against A. cylindrica, and formed alginase, caseinase, lipase, fucodian hydrolase, and laminarinase. Moreover, glycosidases for example ¥â-galatosidase, ¥â-glucosidase, ¥â-glucosaminidase, and ¥â-xylosidase, which hydrolyzed ¥â-O-glycosidic bonds, were found in cell-free extracts of A. johnsonii AK-07. Other glycosidase such as ¥á-galactosidases, ¥á-N-Ac-galctosidases, ¥á-mannosidases, and ¥á-L-fucosidases, which cleavage ¥á-O-glycosidic bonds are not detected. In the results, enzyme systems of A. johnsonii AK-07 were very complex to degrade cell walls of cyanobacteria. The polysaccharides or peptidoglycans of A. cylindrica may be hydrolyzed and metabolized to a range of easily utilizable monosaccharides or other low molecular weight organic substances by strain AK-07 of A. johnsonii. |
